No, Asclepius could not have been substituted for Demophoön in the Eleusinian Mysteries. Chronologically this would have been impossible, if we accept the story that Asclepius lived after the Mysteries had already been established. Otherwise how could he have come to Eleusis with the express intention of being initiated?

Then again, logical chronology never was much of a concern for the Greek mythmakers...
